Can I use the modulation input and the current setpoint at the same time?

Yes.

The modulation input is added to the programmed current setpoint.

For example, you can set a DC operating current with the current setpoint and use the modulation input to apply a varying current around that operating point.

Make sure that the resulting instantaneous current remains within the allowed output-current range of the driver and the safe operating range of the load.
